In this study, immobilization of laccase (L) enzyme on magnetite (Fe(3)O(4)) nanoparticles was achieved, so that the immobilized enzyme could be used repeatedly. For this purpose, Fe(3)O(4) nanoparticles were coated and functionalized with chitosan (CS) and laccase from Trametes versicolor was immobilized onto chitosan-coated magnetic nanoparticles (Fe(3)O(4)-CS) by adsorption or covalent binding after activating the hydroxyl groups of chitosan with carbodiimide (EDAC) or cyanuric chloride (CC). For chitosan-coated magnetic nanoparticles, the thickness of CS layer was estimated as 1.0-4.8 nm by TEM, isoelectric point was detected as 6.86 by zeta (zeta)-potential measurements, and the saturation magnetization was determined as 25.2 emu g(-1) by VSM, indicating that these nanoparticles were almost superparamagnetic. For free laccase and immobilized laccase systems, the optimum pH, temperature, and kinetic parameters were investigated; and the change of the activity against repeated use of the immobilized systems were examined. The results indicated that all immobilized systems retained more than 71% of their initial activity at the end of 30 batch uses. (C) 2011 Wiley Periodicals, Inc. J Appl Polym Sci 123:707-716, 2012

Preparation and Characterization of Amino-Functionalized Zeolite/SiO2 Materials for Trypsin-Chymotrypsin Co-immobilization
Dogan, Demet; Sezer, Selda; Ulu, Ahmet; KÖYTEPE, SÜLEYMAN; ATEŞ, BURHAN (2021-08-01)
Inorganic supports have attracted increased attention in enzyme immobilization since they not only improve enzyme stability but also reduce the final cost of enzymatic reactions. Herein, we explored the suitability of the amino-functionalized zeolite/SiO2 materials to co-immobilize trypsin-chymotrypsin mixture. For this purpose, the trypsin-chymotrypsin mixture was co-immobilized on the amino-functionalized zeolite/SiO2 materials and the immobilization yield was 80.7 +/- 7.6%. The pre-support and its modifi...
Preparation of silica coated cobalt ferrite magnetic nanoparticles for the purification of histidine-tagged proteins
Aygar, Gulfem; Kaya, Murat; Özkan, Necati; Kocabıyık, Semra; Volkan, Mürvet (2015-12-01)
Surface modified cobalt ferrite (CoFe2O4) nanoparticles containing Ni-NTA affinity group were synthesized and used for the separation of histidine tag proteins from the complex matrices through the use of imidazole side chains of histidine molecules. Firstly, CoFe2O4 nanoparticles with a narrow size distribution were prepared in an aqueous solution using the controlled co-precipitation method. In order to obtain small CoFe2O4 agglomerates, oleic acid and sodium chloride were used as dispersants. The CoFe2O4...
Development of enhanced ultrafiltration methodologies for the resolution of racemic benzoin
Olceroglu, Ayse Hande; Çalık, Pınar; Yılmaz, Levent (Elsevier BV, 2008-09-15)
In the scope of achieving the separation of chiral molecules, enzyme enhanced ultrafiltration (EEUF), a new method based on polymer enhanced ultrafiltration (PEUF), utilizing apoenzymes as ligands, was developed. Benzoin was chosen as the model chiral molecule. Bovine serum albumin (BSA) and apo form of benzaldehyde lyase (BAL) (E.C. 4.1.2.38) were used as chiral ligands in PEUF and EEUF experiments, respectively. In order to bind to the target enantiomer well, the addition of ligand to the benzoin solution...

In this study, a novel amperometric glucose biosensor based on a selenium comprising conducting polymer and calixarene was developed. Firstly, poly(2-(2-octyldodecyl)-4,7-di(selenoph-2-yl)-2H-benzo[d][1,2,3]-triazole), poly((SBTz)) was electrodeposited onto a graphite electrode by an electropolymerization technique. Then, a newly synthesized calixarene and gold nanoparticle (AuNP) mixture was used for the improvement of biosensor characteristics. GOx, as a model enzyme was immobilized on the modified electr...
Characterization of extracellular beta-lactamases from penicillin G-resistant cells of Streptococcus thermophilus
Chirica, LC; Güray, Nülüfer Tülün; Gültekin, Güzin Candan; Bozoglu, F (International Association for Food Protection, 1998-07-01)
In this study, biochemical properties of two extracellular beta-lactamases produced by penicillin-resistant Streptococcus thermophilus cells were investigated. Both beta-lactamases showed specificity for penicillins but not for cephaloridins. The p-lactamases exhibited different affinities for penicillin G. The one with the higher molecular weight (F1) had a K(m) value of 3.44 mu M and a V(max), value of 8.33, mu mol/min/mg of protein, whereas the beta-lactamase with the lower molecular weight (FII) had a K...
