Collagen-based micropatterned films were seeded with human corneal keratocyte and epithelial cells to study their mechanical properties as tissue engineering substrates. The patterns were in the form of parallel channels with slanted walls. Influence of cell presence, type and growth on the mechanical properties of the films was investigated. Unseeded films showed gradual strength reduction from an initial value of 0.046 N/mm, possibly due to degradation, down to 0.032 +/- 0.012 N/mm in 2 weeks. Keratocyte growth was found to significantly improve the mechanical behavior of the films upon 1 week of incubation (0.067 +/- 0.017N/mm) and the improvement continued gradually over the next 2 weeks. Films seeded with D407 retinal pigment epithelial cells, on the other hand, experienced a decrease (0.023 +/- 0.011 N/mm), followed by a slight increase in mechanical properties in the 21-day period. A steady increase in the number of keratocytes along the channels, cytoskeleton alignment and extracellular matrix (ECM) secretion restricted to the channels was observed. Increase in strength observed with keratocytes and, to a lesser extent, with the epithelial cells can be attributed to directional ECM synthesis and the orientation of the cells and their cytoskeleton which contribute to the strength in the direction of the channels. This study showed that cell, especially keratocyte, presence compensates for the degradation of collagen films and improve the overall mechanical properties of the engineered tissue.

Influence of keratocytes and retinal pigment epithelial cells on the mechanical properties of polyester-based tissue engineering micropatterned films
Zorlutuna, Pinar; Builles, Nicolas; Damour, Odile; Elsheikh, Ahmed; Hasırcı, Vasıf Nejat (2007-08-01)
In this paper the mechanical properties of micropatterned polyester films prepared to serve as tissue engineering scaffolds of cornea were examined. Films were prepared by solvent casting of blends of poly(L-lactide-co-D,L-lactide) and poly(3-hydroxybutyric acid-co-3-hydroxyvaleric acid), on a micropatterned silicon template. They were seeded with keratocytes or retinal pigment epithelia] cells and subjected to tensile testing to assess the contribution of cells and the deposited extra-cellular matrix (ECM)...
Influence of nanopatterns on endothelial cell adhesion: Enhanced cell retention under shear stress
Zorlutuna, P.; Rong, Z.; Vadgama, P.; Hasırcı, Vasıf Nejat (2009-09-01)
In this study, nanopatterned crosslinked films of collagen Type I were seeded with human microvascular endothelial cells and tested for their suitability for vascular tissue engineering. Since the films will be rolled into tubes with concentric layers of collagen, nutrient transfer through the collagen films is quite crucial. Molecular diffusivity through the collagen films, cell viability, cell proliferation and cell retention following shear stress were studied. Cells were seeded onto linearly nanogrooved...
Contact guidance enhances the quality of a tissue engineered corneal stroma
Vrana, E.; Builles, N.; Hindie, M.; Damour, O.; AYDINLI, ATİLLA; Hasırcı, Vasıf Nejat (2008-02-01)
Corneal stroma is a very complex structure, composed of 200 lamellae of oriented collagen fibers. This highly complex nature of cornea is known to be important for its transparency and mechanical integrity. Thus, an artificial cornea design has to take into account this complex structure. In this study, behavior of human corneal keratocytes on collagen films patterned with parallel channels was investigated. Keratocytes proliferated well on films and reached confluency after 7 days in the incubation medium....
Effects of silica nanoparticles on the performance of water-based drilling fluids
Kök, Mustafa Verşan; Bal, Berk (2019-09-01)
In this research, two groups of experiments were conducted to investigate the effects of silica (SiO2) nanoparticles on the filtration and rheological properties of water-based drilling fluids. In the first group, bentonite, chrome-free lignosulfonate (CFL) and carboxymethyl cellulose (CMC) were used in different concentrations to obtain base fluids. Nanofluids were prepared by adding 0.5 g of four different silica nanoparticles into these drilling fluids. Comparison of rheological properties, fluid loss am...
Effect of boron containing materials on flammability and thermal degradation of polyamide-6 composites containing melamine cyanurate
Yılmaz, Ayşen; Bayramli, Erdal (2011-05-01)
Three different boron containing materials, zinc borate (ZnB), borophosphate (BPO(4)), and boron and silicon containing oligomer (BSi), were used to improve the flame retardancy of melamine cyanurate (MC) in a polyamide-6 (PA-6) matrix. The combustion and thermal degradation characteristics were investigated using limiting oxygen index (LOI), UL-94 standard, thermogravimetric analysis-Fourier transform infrared spectroscopy (TGA-FTIR), differential scanning calorimeter (DSC), and scanning electron microscop...
