In the present work, mutual interaction of melittin, a pore forming hemolytic toxin from bee venom, and vitamin D-2, an antioxidant steroid, with dipalmitoyl phosphatidylcholine (DPPC) multilamellar liposomes has been investigated. Turbidity and Fourier transform infrared (FTIR) spectroscopic measurements, in combination with thermodynamic calculations, were used to monitor the modulating effect of vitamin D-2 on a melittin-DPPC membrane system. The results indicate that melittin on its own decreases the main phase transition to lower temperatures and also dramatically decreases the stability of the membrane. It has an overall disordering effect on the phospholipid membrane structures. Inclusion of vitamin D-2 at low concentrations (3, 6 mol%) into melittin containing DPPC liposomes slightly shifts the main phase transition to lower temperatures. High concentration of vitamin D-2 (9 mol%) has a more dramatic effect in shifting the main phase transition to lower temperature. It also causes a significant broadening in the phase transition curve. The present study also demonstrates that, with the addition of vitamin D-2 into melittin-DPPC system, absorbance value in turbidity study and the frequency of the CH, stretching band in FTIR study changes in a manner that are consistent with a reduction in the membrane perturbing effect of melittin on DPPC liposomes. Vitamin D-2 diminishes the disordering effect of melittin on DPPC lipids and produces a more ordered membrane system. These results were confirmed with thermodynamic calculations.

Turbidity technique was employed to study the mutual interaction of melittin and vitamin D-2 With 1,2-dipalmitoyl phosphatidylcholine (DPPC) bilayers. Ln addition, the data obtained from the phase transition curve is used to calculate the main phase transition temperature, conformational stability and activation energy values by using two state transition thermodynamic approach. The results indicate that melittin decreases the main phase transition temperature and also dramatically decreases the stability. ...
Vitamin E Derivative with Modified Side Chain Induced Apoptosis by Modulating the Cellular Lipids and Membrane Dynamics in MCF7 Cells

The vitamin E derivative with side chain modification (TC6OAc) has been shown to possess anticancer activity in our earlier in vivo studies. It was hypothesized that, as Vitamin E (VE) and VE derivative are fat soluble lipophilic molecules, they exert their function by modulating the lipid metabolism and related pathways. This study aimed to evaluate the cellular impact of this VE derivative (2,5,7,8-Tetramethyl-2-(4 '-Methyl-3 '-Pentenyl)-6-Acetoxy Chromane-TC6OH), using alpha-tocopherol as a reference com...

Alkaline phosphatase (ALP), an enzyme involved in mineralization of bone, is incorporated into three hydrogel biomaterials to induce their mineralization with calcium phosphate (CaP). These are collagen type I, a mussel-protein-inspired adhesive consisting of PEG substituted with catechol groups, cPEG, and the PEG/fumaric acid copolymer OPF. After incubation in Ca-GP solution, FTIR, EDS, SEM, XRD, SAED, ICP-OES, and von Kossa staining confirm CaP formation. The amount of mineral formed decreases in the orde...
Squaramide catalyzed alpha-chiral amine synthesis
Karahan, SEDA; Tanyeli, Cihangir (2018-10-17)
Enantiomerically pure alpha-chiral amines, have been commonly utilized as resolving agents and chiral auxiliaries and are currently found in 40% of active pharmaceutical ingredients. Hence, development of highly stereoselective metal-free protocols regarding atom-economy and large-scale applications becomes a major issue. In this respect, chiral bifunctional H-bonding squaramides have been successfully applied for both amine synthesis and functionalization of amines in the last decade. This survey summarize...

Botulinum neurotoxins (BoNTs), the causative agents of botulism, are potent inhibitors of neurotransmitter release from motor neurons. There are currently no drugs to treat BoNT intoxication after the onset of the disease symptoms. In this study, we explored how modulation of key host pathways affects the process of BoNT intoxication in human motor neurons, focusing on Src family kinase (SFK) signaling. Motor neurons derived from human embryonic stem (hES) cells were treated with a panel of SFK inhibitors a...
