In this study, the performance of a two stage rotating biological contactor (RBC) was evaluated for the treatment of synthetic wastewater containing peptone, 4-chlorophenol (4-CP) and 2,4-dichlorophenol (2,4-DCP) at 5 rpm. Also, the effect of biogenic substrate (peptone) concentration on the reactor performance was investigated. High chlorophenols (> 98%) and COD (> 94%) removals were achieved throughout the reactor operation in the first stage and the second stage behaved as a polishing step. The observed max [mum 4-CP and 2,4-DCP removal rates in the first stage were 2305 mg/L day (18.3 g/m(2) day) and 1202 mg/L day (9.5 g/m(2) day), respectively. Deacclimation of biomass was carried out for 3.5 months after 260 days of operation with the reactor feed turning on to contain peptone only. Upon addition of 4-CP (200 mg/L) and 2,4-DCP (100 mg/L), a rapid reacclimation of biomass was observed within 16 days. Shock loading experiments with 4-CP (822.7 +/- 1.4) and 2,4-DCP (424.6 +/- 1.9 mg/L) resulted in around four times higher effluent 4-CP concentrations than 2,4-DCP in both stages, which may be due to strong competitive inhibition of 2,4-DCP on 4-CP degradation. Covalent immobilization of chloroperoxidase onto magnetic beads: Catalytic properties and stability
Bayramoglu, Guelay; Kiralp, Senem; Yilmaz, Meltem; Toppare, Levent Kamil; Arica, M. Yakup (Elsevier BV, 2008-02-15)
Amino groups containing magnetic beads were used in covalent immobilization of the enzyme "chloroperoxidase (CPO)" which is one of a few enzymes that can catalyse the peroxide dependent oxidation of a wide spectrum of organic and inorganic compounds. The magnetic poly(glycidylmethacrylate-methylmethacrylate-etbyleneglycol dimethacrylate), magnetic p(GMA-MMA-EGDMA) beads were prepared via suspension polymerization in the presence of ferric ions. The magnetic beads were characterized with scanning electron mi...
The biodegradation of benzene, toluene and phenol in a two-phase system
Hamed, TA; Bayraktar, E; Mehmetoglu, U; Mehmetoglu, T (Elsevier BV, 2004-07-15)
A two-phase aqueous-organic system was used to degrade benzene, toluene and phenol, individually and as mixture by Pseudomonas putida F1 (ATCC 700007). In the first stage of the work, the effect of the phase ratio (0-1, v/v) and the agitation rate (150 and 200 rpm) on the biodegradation process was investigated in an orbital shaker using 2-undecanone as the solvent and the most suitable phase ratio and agitation rate were found to be 0.0625 (v/v) and 200 rpm, respectively. P. putida F1 was added into the aq...
Glucose isomerase production on a xylan-based medium by Bacillus thermoantarcticus
Çalık, Pınar; Haykir, Nazife Isik; BOYACI, İSMAİL HAKKI (Elsevier BV, 2009-01-15)
Effects of medium components on intracellular glucose isomerase (GI) production were investigated by Bacillus thermoantarcticus. The highest GI activity was obtained as 1630 U dm(-3) in the medium containing (g dm(-3)): 10.6, birchwood-xylan: 5.6, yeast extract: 5.9 (NH(4))(2)SO(4) at T = 55 C in 33 cm(-3) shake-flasks. When birchwood-xylan was replaced with oat spelt- or beechwood-xylan, GI activity decreased to 1372 and 1308 U dm(-3). respectively. Effects of pH at uncontrolled-pH (pH(UC) = 6.0) and contr...
QCM-based DNA biosensor for detection of genetically modified organisms (GMOs)
Karamollaoglu, Irem; Oektem, Hueseyin Avni; Mutlu, Mehmet Kayhan (Elsevier BV, 2009-05-15)
Development of a mass sensitive quartz crystal microbalance (QCM)-based DNA biosensor for the detection of the hybridization of CaMV 35S promoter sequence (P35S) was investigated for the screening of genetically modified organisms (GMOs). Attention was focused on the choice of the coating chemistry that could be used for the immobilization of probe sequences on the gold surface of the quartz crystal. Two immobilization procedures were tested and compared considering the amount of the immobilized P35S probe ...
Carbon sources affect metabolic capacities of Bacillus species for the production of industrial enzymes: theoretical analyses for serine and neutral proteases and alpha-amylase
Çalık, Pınar (Elsevier BV, 2001-07-01)
The metabolic fluxes through the central carbon pathways were calculated for the genus Bacillus separately for the enzymes serine alkaline protease (SAP), neutral protease (NP) and alpha -amylase (AMY) on five carbon sources that have different reduction degrees (gamma), to determine the theoretical ultimate limits of the production capacities of Bacillus species and to predict the selective substrate for the media design. Glucose (gamma = 4.0), acetate (gamma = 4.0), and the TCA cycle organic-acids succina...
