Synthetic ODNs expressing CpG motifs trigger an innate immune response via TLR9. pDCs are major effectors of this response. Two structurally distinct classes of CpG ODNs have been identified that differentially activate pDCs. "K" ODNs trigger the production of TNF-alpha and IL-6, whereas "D" ODNs preferentially induce the secretion of IFN-alpha. As K and D ODNs have distinct therapeutic effects, knowledge of their shared and sequence-specific activity is of considerable importance. This work uses the CAL-1 human pDC line to analyze the effect of CpG stimulation on gene expression. Genes up-regulated by both K and D ODNs (n = 92) were largely dependent on type I IFN signaling and characterized functionally by antiviral activity. K ODNs induced a short-term increase in IFN-alpha/beta production and uniquely up-regulated genes that supported antibacterial responses. In contrast, D ODNs triggered a persistent increase in IFN-alpha/beta production and uniquely up-regulated genes associated with metabolic functions. Thus, the core functionality of human pDCs mediated by TLR9 ligation rests on a type I IFN response that differs from the response induced by the structural elements unique to specific classes of ODNs. Regulation of CpG-induced immune activation by suppressive oligodeoxynucleotides.
Klinman, DM; Zeuner, R; Yamada, H; Gürsel, Mayda; Currie, D; Gursel, I (Wiley, 2003-12-01)
Bacterial DNA and synthetic oligodeoxynucleotides (ODN) containing unmethylated "CpG motifs" stimulate an innate immune response characterized by the production of cytokines, chemokines, and polyreactive Igs that promote host survival following infectious challenge. Yet CpG-driven immune activation can have deleterious consequences, such as increasing the host's susceptibility to autoimmune disease. The immunomodulatory activity of CpG DNA can be blocked by DNA containing "suppressive" motifs. This work exp...
Human peripheral blood cells differentially recognize and respond to two distinct CpG motifs
Verthelyi, D; Ishii, KJ; Gürsel, Mayda; Takeshita, F; Klinman, DM (The American Association of Immunologists, 2001-02-15)
Oligodeoxynucleotides (ODN) that contain unmethylated CpG dinucleotides trigger a strong innate immune response in vertebrates. CpG ODN show promise as vaccine adjuvants, anti-allergens, and immunoprotective agents in animal models. Their transition to clinical use requires the identification of motifs that are optimally stimulatory in humans. Analysis of hundreds of novel ODN resulted in the identification and characterization of two structurally distinct "clusters" of immunostimulatory CpG ODN, One cluste...
Differential and competitive activation of human immune cells by distinct classes of CpG oligodeoxynucleotide
Gürsel, Mayda; Gursel, I; Ishii, KJ; Klinman, DM (2002-05-01)
Synthetic oligodeoxynucleotides (ODN) expressing "CpG motifs" show promise as immune adjuvants, antiallergens, anticancer, and immuno-protective agents. Two structurally distinct classes of CpG ODN have been identified that stimulate human PBMC. This work establishes that both types of ODN bind to and are internalized by the same individual B cells, NK cells, and monocytes. However, the intracellular localization of "D" and "K" ODN differs, as does their functional activity: "K" type ODN trigger monocytes a...
Alterations of ceramide/sphingosine 1-phosphate rheostat involved in the regulation of resistance to imatinib-induced apoptosis in K562 human chronic myeloid leukemia cells
Baran, Yusuf; Salas, Arelis; Senkal, Can E.; Gündüz, Ufuk; Bielawski, Jacek; Obeid, Lina M.; Ogretmen, Besim (2007-04-13)
In this study, mechanisms of resistance to imatinib-induced apoptosis in human K562 cells were examined. Continuous exposure to stepwise increasing concentrations of imatinib resulted in the selection of K562/IMA-0.2 and -1 cells, which expressed similar to 2.3- and 19-fold resistance, respectively. Measurement of endogenous ceramides by high performance liquid chromatography/mass spectroscopy showed that treatment with imatinib increased the generation of ceramide, mainly C-18-ceramide, which is generated ...
Enzyme catalyzed hydroxymethylation of aromatic aldehydes with formaldehyde. Synthesis of hydroxyacetophenones and (S)-benzoins
Demir, Ayhan Sıtkı; Ayhan, P; Igdir, AC; Duygu, AN (Elsevier BV, 2004-07-26)
Benzaldehyde lyase from the Pseudomonas Fluorescens catalyzed reaction of aromatic aldehydes with formaldehyde providing 2-hydroxy-1-arylethan-1-one in high yields via an acyloin linkage. Kinetic resolution of rac-benzoins with formaldehyde providing (S)-benzoins and 2-hydroxy-1-arylethan-1-one via C-C bond cleavage and a bond formation reaction.
