Beet molasses based exponential feeding strategy for thermostable glucose isomerase production by recombinant Escherichia coli BL21 (DE3)
Angardi, Vahideh; Çalık, Pınar (Wiley, 2013-05-01)
BACKGROUND: The effects of pretreated beet molasses based feeding strategies on thermostable glucose isomerase (GI) production by recombinant Escherichia coli BL21 (DE3) pLysS were investigated. RESULTS: The thermostable GI encoding gene of Thermus thermophilus (xylA) was recombined with pRSETA vector, and the pRSETA::xylA obtained was transferred into E.coli BL21 (DE3) pLysS and used for GI production. The highest soluble GI activity was obtained at t = 30 h, as A = 16 400 U L1 (20.6 U mg1 protein) under m...
Enzymatic Hydrolysis of Fruit Peels and Other Lignocellulosic Biomass as a Source of Sugar
POCAN, Pelin; BAHÇEGÜL, Erinc; Öztop, Halil Mecit; Hamamcı, Haluk (2018-06-01)
Glucose and reducing sugars were obtained from corn cobs, orange and pomegranate peels by employing different enzymatic hydrolysis and pretreatment strategies. For the hydrolysis of corn cobs, effect of alkaline pretreatment was examined. Effect of cellobiase loading on hydrolysis was also investigated. Glucose content was measured using a Blood Glucose Monitor. When cellobiase was also added to hydrolysate, glucose and TRS yield increased from 20 and 34% respectively. Unlike corncobs, alkaline pretreatment...
Optimising clarification of carrot juice by bacterial crude pectinase
Uzuner, Sibel; Çekmecelioğlu, Deniz (2015-12-01)
This study was undertaken to search for potential use of crude bacterial pectinase enzyme produced from Bacillus subtilis grown on hazelnut shell hydrolysate in clarification of carrot juice and to optimize the enzyme load, pH and time using the Box-Behnken response surface methodology (RSM). The carrot juice was treated with the crude pectinase enzyme (5.60 U mL(-1)) at different concentrations (0.1-0.5%), pH (4-7), and time (2-6 h). The obtained enzyme was also compared with commercial fungal pectinase at...
Biological hydrogen production from sugar beet molasses by agar immobilized R. capsulatus in a panel photobioreactor
ELKAHLOUT, Kamal; SAĞIR, EMRAH; ALIPOUR, Siamak; Koku, Harun; Gündüz, Ufuk; EROĞLU, İNCİ; Yücel, Ayşe Meral (2018-03-18)
Biohydrogen production from sugar beet molasses was investigated by using agar immobilized R. capsulatus YO3. A panel photobioreactor (1.4 L) was employed for a long-term hydrogen production in both indoor and outdoor conditions. The impact of several initial molasses concentrations on hydrogen production, yield and productivity were assessed. Indoor studies revealed that initial sucrose concentration in molasses should be kept below 20 mM to prevent inhibition of hydrogen production. The highest hydrogen p...
Anaerobic acidification of sugar-beet processing wastes: Effect of operational parameters
Alkaya, Emrah; Demirer, Göksel Niyazi (Elsevier BV, 2011-01-01)
The objective of this study was to maximize the hydrolysis and acidification of sugar-beet processing wastewater and beet pulp for volatile fatty acid (VFA) production through acidogenic anaerobic metabolism. Experiments were conducted to determine the optimum operational conditions (HRT, waste-mixing ratio and pH) for effective acidification in daily-fed, continuously mixed anaerobic reactors. For this purpose, reactors were operated at 35 +/- 1 degrees C with different combinations of HRT (2-4 days), wast...
