In order to understand the conformational changes of polyphenol oxidase (PPO), which is a food quality related enzyme, after thermal treatment, secondary structure changes of the enzyme were analyzed by using Fourier Transform Infrared (FTIR) spectroscopy and compared with the change in enzyme activity in the temperature range of 25-80 degrees C. Fourier self-deconvolution, neural network (NN) and curve-fitting analysis were applied to the amide I band of FTIR spectra for detail analysis of secondary structure elements. FTIR analysis indicated that PPO is an a-helix dominating enzyme. Detail analysis revealed that, as temperature increased, alpha-helix and beta-sheet decreased, but aggregated beta-sheet, turns and random coil increased. The marked changes were noted at 40 degrees C with the occurrence of new bands due to aggregated beta-sheet structures, all of which indicate protein denaturation. These aggregation bands were still observed when the temperature was reduced back to 25 degrees C, from 70 degrees C, demonstrating an irreversible change in the structure.

Secondary structure and conformational change of mushroom polyphenol oxidase during thermosonication treatment by using FTIR spectroscopy
Baltacıoğlu, Hande; Bayındırlı, Alev; Severcan, Feride (2017-01-01)
To understand the conformational changes of mushroom PPO, the secondary structural change of the enzyme during thermosonication treatment at different power (60, 80 and 100%), temperature (20-60 degrees C) and time (0-30 min) combinations was investigated by using FTIR spectroscopy and compared with the change in enzyme activity. The enzyme inactivation higher than 99% was obtained at 100% amplitude at 60 degrees C for 10 min. FTIR studies showed that marked spectral changes were noted after ultrasound trea...
Effects of the non-steroidal anti-inflammatory drug celecoxib on cholesterol containing distearoyl phosphatidylcholine membranes
Sade, Asli; Banerjee, Sreeparna; Severcan, Feride (2011-01-01)
The effects of different concentrations of celecoxib on the acyl chain order, dynamics and the hydration status of the head group and interfacial region of model membranes containing DSPC and cholesterol were investigated in detail using Fourier transform infrared spectroscopy. Our results reveal that regardless of the presence of cholesterol, celecoxib is able to alter the physical properties of membranes. It exerts opposing effects on membrane order at high and low concentrations and decreases membrane fl...
Effect of gramicidin S on the dipalmitoylphosphatidyl-glycerol thermotropic phase transition in DPPG/GS systems: A mathematical approach
Stan, Cristina; Cristescu, C. P.; Severcan, Feride; Dorohoi, Dana (Informa UK Limited, 2006-01-01)
We present a mathematical approach to the experimental data recorded via Fourier transform-infrared spectroscopy regarding the influence of the concentration of the antimicrobial peptide gramicidin S (GS) on the thermotropic phase transition of the dipalmitoylphosphatidyl-glycerol (DPPG) lipid bilayer membrane in DPPG/GS systems. The model is based on the influence of the GS concentration on the parameters of a nonlinear damped oscillator, which models the CH2 symmetric stretching band.
Effect of silver encapsulation on the local structure of titanosilicate ETS-10
GALIOGLU, Sezin; ZAHMAKIRAN, Mehmet; Kalay, Yunus Eren; Özkar, Saim; Akata, Burcu (2012-09-01)
Silver(0) nanoparticles stabilized by titanosilicate (ETS-10) framework were synthesized by following a simple two step procedure involving the incorporation of silver(I) (Ag+) ions into ETS-10 matrix via ion-exchange with extra framework Na+ and K+ cations followed by their reduction with sodium borohydride (NaBH4) in aqueous medium all at room temperature. Silver(0) nanoparticles dispersed in the ETS-10 matrix were collected as gray powders and characterized by using advanced analytical methods including ...
Effect of progesterone on DPPC membrane: Evidence for lateral phase separation and inverse action in lipid dynamics
Korkmaz, F; Severcan, Feride (Elsevier BV, 2005-08-15)
Interactions of progesterone with zwitterionic dipalmitoyl phosphatidylcholine (DPPC) triultilamellar liposomes were investigated as a function of temperature and progesterone concentration by using three non-invasive techniques namely Fourier transform infrared spectroscopy, turbidity at 440 nm, and differential scanning calorimetry. The results reveal that progesterone changes the physical properties of DPPC bilayers by decreasing the main phase-transition temperature, abolishing the pre-transition, broad...
