An aqueous slurry of gamma-irradiated sterilized dried figs was inoculated with toxigenic strains of Aspergillus flavus and Aspergillus parasiticus. During incubation at 28 degrees C, pH, fructose, glucose, and free amino acids were determined by high-performance liquid chromatography and liquid chromatography (LC)/mass spectrometry, respectively, over 13 time points (1-20 days). At the same 13 time points using a LC/time-of-flight mass spectrometry screening method, aflatoxin B, and other secondary metabolites were simultaneously monitored. During the course of incubation, the pH significantly decreased and aflatoxin B, formation correlated with a reduction in proline content for both fungi. Of the 22 free amino acids that were monitored, only proline and cystine were found to be critical in supporting aflatoxin production. Levels of fructose and glucose steadily declined during incubation, until glucose was almost exhausted after 21 days. These time-course experiments confirmed the need for carbon and nitrogen sources for aflatoxin production in dried figs, and the favorable composition of figs as a fungal growth medium.

Analysis of cellulase and polyphenol oxidase production by southern pine beetle associated fungi
Valiev, Abduvali; Ögel, Zümrüt Begüm; Klepzig, Kier D. (Springer Science and Business Media LLC, 2009-09-01)
In this study, the production of extracellular enzymes by fungi associated with southern pine beetle was investigated for the first time. Cellulase and polyphenol oxidase production were analyzed for three beetle associated fungi. Only the mutualistic symbiont Entomocorticium sp. A was found to produce cellulases and polyphenol oxidase. In time course analyses of cellulase production in batch cultures, Entomocorticium sp. A showed maximum activity of 0.109 U/ml and 0.141 U/ml for total cellulase and endoglu...
Optimization of solid-liquid extraction of resveratrol and other phenolic compounds from milled grape canes (Vitis vinifera)
Karacabey, Erkan; Mazza, Giuseppe (American Chemical Society (ACS), 2008-08-13)
Optimization of the solid-liquid extraction conditions for trans-resveratrol, trans-E-viniferin, ferulic acid, and total phenolics from milled grape canes has been investigated. The temperature and ethanol concentration were found to be major process variables for all responses, whereas the solvent to solid ratio was found not to be significant for any of the responses studied. The yields of transresveratrol, trans-epsilon-viniferin, and total phenolics increased with increasing temperature. Maximum yields ...
SUPERCRITICAL-FLUID EXTRACTION OF CARBOXYLIC AND FATTY-ACIDS FROM AGARICUS SPP MUSHROOMS
ABDULLAH, MI; YOUNG, JC; GAMES, DE (American Chemical Society (ACS), 1994-03-01)
Material obtained by the CO2 supercritical fluid extraction (SFE) of the wild and cultivated mushrooms Agaricus silvicola and Agaricus bisporus, respectively, was analyzed by g chromatography (GC)-mass spectrometry and GC-Fourier transform infrared spectrometry. SFE gave results comparable with classical extraction techniques for free fatty acids and esters but with fewer manipulative steps and also resulted in the characterization of acids not previously reported from these sources. The organic acids ident...
Utilization of pretreated molasses for serine alkaline protease production with recombinant Bacillus species
Calik, G; Pehlivan, N; Kalender, N; Ozdamar, TH; Çalık, Pınar (Informa UK Limited, 2003-05-01)
Recombinant Bacillus amyloliquefaciens, Bacillus cereus, Bacillus subtilis , and Bacillus licheniformis were used for the production of serine alkaline protease (SAP) utilizing chemically and/or physically pretreated molasses. The highest enzyme activity was obtained with r- Bacillus subtilis , with the complex medium involving physically treated molasses having 20 kg m(-3) initial sucrose concentration in small-scale, agitation- and heating rate-controlled bioreactors at t=63 h. Effects of oxygen transfer ...
DETERMINATION OF SELENIUM IN DIET BY ZEEMAN EFFECT GRAPHITE-FURNACE ATOMIC-ABSORPTION SPECTROMETRY FOR CALCULATION OF DAILY DIETARY INTAKES
ARI, U; Volkan, Mürvet; ARAS, NK (American Chemical Society (ACS), 1991-12-01)
The selenium content of Turkish diets was determined by Zeeman effect graphite furnace atomic absorption spectrometry (GFAAS). Samples, collected from two different socioeconomic groups by duplicate portion technique, were digested with concentrated nitric acid in acid bombs, and hydrogen peroxide was added for complete oxidation. The digested solutions were then analyzed by GFAAS with the nickel chemical modifier addition. The detection limit for Se(VI) was 10.5 ng/mL for aqueous standard solutions. Averag...
