Bu çalışmada termofilik bir küf olan Thermomyces lanigunosus'un hücre dışı polifenol oksidaz üretimi incelenmiş, çeşitli besin kaynaklarının, indükleyici bazı maddelerin ve değişik fermantasyon parametrelerinin (pH, sıcaklık, çözünmüş oksijen konsantrasyonu) enzim üretimi üzerine etkisi araştırılmış, enzim kısmı olarak saflaştırılıp karakterize edilmiştir. Thermomyces lanuginosus'a. göre daha yüksek polifenol oksidaz üretimi gerçekleştirebilen Scytalidium thermophilum polifenol oksidazları ise kolon kromatografi ile saflaştırıp karakterize edilmiştir. Biyokimyasal karakterizasyon için pH ve ısıl dayanıklılık, pH ve sıcaklığın enzim aktivitesi üzerine etkisi, kinetik sabitleri, molekül ağırlığı ve izoelektrik noktası tespit edilmiştir.
