Glutathione S-transferases in breast tissue play an important role in the susceptibility to the mutagenic effects of chemical carcinogens and in the response of breast tumors to chemotherapy. In this study the immunohistochemical staining characteristics of glutathione S-transferase isoenzymes (alpha, mu, pi, and theta) were investigated in invasive duct carcinomas and in normal breast tissue of 43 patients. The relationships between the expression of the GST isoenzymes and some clinicopathological features were also examined. Diffuse cytoplasmic staining of varying intensity was observed for GST alpha, theta, and pi in normal and tumorous breast tissue in 100% of the samples. In normal epithelium there was a stronger intensity of staining for GST alpha, mu. and pi expression than in invasive tumor tissues (P 0.05). In this study significant relationships were observed between microcalcification status and GST mu, between menopause status and GST alpha, and between tumor grade and GST mu expression (P 0.05).

Development and investigation of etoposide resistance in MCF-7 Breast cancer cell line
Kaplan, Esra; Gündüz, Ufuk; Department of Biology (2010)
Failure of chemotherapy in cancer patients because of development of drug resistance is a major problem. Alterations of DNA repair mechanisms and drug targets are among the important resistance mechanisms which are developed against topoisomerase II inhibitors etoposide and doxorubicin. Modifications in the expression levels of mismatch repair (MMR) genes due to resistance to topoisomerase II inhibitors are involved in breast cancer. In this study, etoposide resistant sublines were developed from MCF7 breas...
Implementation of a fast simulation tool for the analysis of contrast mechanisms in HMMDI and enhancement of the SNR in the experimental set-up
İrgin, Ümit; Gençer, Nevzat Güneri; Top, Can Barış; Department of Electrical and Electronics Engineering (2021-9-06)
Clinical method for breast tumor detection is Mammography (X-rays), which have limitations and may yield inaccurate results. Alternative novel techniques are required to characterize the breast tissues and extract accurate information for identification of malignancies in the tissue. Harmonic Motion Microwave Doppler Imaging (HMMDI), which enhances hybridizing microwave signals and ultrasound techniques, has been recently proposed for detection of tumors in the tissue. In HMMDI method, the data is a combina...
Investigation of the lipid metabolic changes induced by novel alpha tocopherol analog (TC6OH) in luminal a type breast cancer cells
Gök, Seher; Severcan, Feride; Department of Biology (2017)
Although the traditional chemotherapeutic agents able to suppress breast tumor progression, they are mostly unspecific in their mechanism of action and have toxic effects to normal tissues. Instead, it has been reported that tumor cells are more sensitive to some forms of Vitamin E (VE) and its synthetic derivatives than healthy cells. Studies of drug-cell interactions and understanding the mechanism underlying cytotoxic activity and biological effects at molecular level in cancer model cells are essential ...
Application of image enhancement algorithms to improve the visibility and classification of microcalcifications in mammograms
Akbay, Cansu; Gençer, Nevzat Güneri; Gençer, Gülay; Department of Biomedical Engineering (2015)
Breast cancer is the second leading cause of cancer deaths for women. Mammography is the most effective technology presently available for breast cancer screening, despite the fact that there are still some limitations of the imaging technique, such as insufficient resolution, low local contrast and noise combined with the subtle nature of the usual radiographic findings. One of the most important radiographic findings associated to the existence of breast cancer is the clustered microcalcifications. Especi...
Comparison of NAT1, NAT2 & GSTT2-2 activities in normal and neoplastic human breast tissues
Geylan-SU, YS; Isgor, B; Coban, T; Kapucuoglu, N; Aydintug, S; Iscan, M; Iscan, M; Güray, Nülüfer Tülün (American Society for Pharmacology & Experimental Therapeutics (ASPET), 2006-01-01)
In this study, arylamine N-acetyltransferases, NATs (E.C.2.3.1.5) and glutathione-S-transferase-T2-2, GSTT2-2 (E.C.2.5.1.18) enzyme activities in the breast tumor and surrounding tumor-free tissues of 22 female breast cancer patients with infiltrating ductal carcinoma were measured. The possible impacts of grade of malignancy, chemotherapy treatment, estrogen receptor status and menopausal status on all enzyme activities were evaluated. The results showed that, both NAT2 and GSTT2-2 display significant diff...
