There are five (A-E) identified mitochondria, DNA (mtDNA) haplogroups (HPGs) in domestic sheep. In search of a quick and reliable HPG classification, three different molecular approaches were tested: the restriction fragment length polymorphism (RFLP) method applied to mtDNA control region (CR), the single strand conformational polymorphism (SSCP) method applied to NADH dehydrogenase subunit 2 (MT-ND2) and to NADH dehydrogenase subunit 4 (MT-ND4) regions. The results were evaluated comparatively to elucidate their power in discriminating the sheep mtDNA HPGs. For this purpose, the HPGs of 622 domestic sheep all from Turkey, hence from the genetic diversity hot spot of domestic sheep distribution were examined using these three approaches. The HPGs for some of the individuals (n = 240) were also obtained based on CR sequences. The SSCP analysis of the MT-ND2 region (by identifying HPGs as A, B, C/E and D) exhibited higher discrimination power, sensitivity and specificity in HPG classification when compared with the SSCP analysis of MT-ND4 region and RFLP analysis of CR.

Exceptional maternal lineage diversity in brown bears (Ursus arctos) from Turkey
Cilingir, F. Gozde; Peksen, Cigdem Akin; Ambarli, Huseyin; Beerli, Peter; Bilgin, Cemal Can (Oxford University Press (OUP), 2016-02-01)
The genetic diversity and phylogeography of maternal lineages in Ursus arctos Linnaeus, 1758 (the brown bear) have been studied extensively over the last two decades; however, sampling has largely been limited to the northern Holarctic, and was possibly biased towards lineages that recolonized the vast expanses of the north as the Last Glacial Maximum (LGM) ended. Here we report the genetic diversity and phylogeography of U.arctos from Turkey based on 35 non-invasive samples, including five from captive ind...
MOLECULAR PHYLOGENY OF SECTION DROSANTHE (SPACH) ENDL. (HYPERICUM L.)INFERRED FROM CHLOROPLAST GENOME
Dogan, Gulden; Yılmaz, Aslı; Bagci, Eyup; Kaya, Zeki (2017-12-01)
The phylogenetic relationships of Drosanthe section of Hypericum genus (Hypericaceae) were analyzed by using non-coding chloroplast DNA region (trnL 3'-trnF) for 58 individuals. The section is represented by 23 taxa and nine of which are endemic to Turkey. The chloroplast phylogeny suggested that the members of this section belonged to a polyphyletic group, which imply at least two independent origins. The individuals of this section clearly formed two main clades. One clade included all members of this sec...
A comparative genetic analysis of the subterranean termite genus Reticulitermes (Isoptera : Rhinotermitidae)
Austin, JW; Szalanski, AL; Uva, P; Bagneres, AG; Kence, Aykut (Oxford University Press (OUP), 2002-11-01)
DNA sequencing analysis of the mitochondrial DNA cytochrome oxidase H (COII) region was used to examine genetic variation in the termite genus Reticulitermes Holmgren. We examined 21 species and subspecies from three continents. Sequencing of a 677-bp region of a 780-bp amplicon from 41 individuals and from 17 sequences obtained from GenBank revealed 221 polymorphic sites within the genus. Tajima-Nei distances from species ranged from 0.9 to 12.7%, and parsimony and maximum likelihood analysis revealed seve...
Pulsed-Field Gel Electrophoresis Diversity of Human and Bovine Clinical Salmonella Isolates
Soyer, Yeşim; Schoonmaker-Bopp, Dainna J.; Root, Timothy P.; Warnick, Lorin D.; McDonough, Patrick L.; Dumas, Nellie B.; Groehn, Yrjo T.; Wiedmann, Martin (Mary Ann Liebert Inc, 2010-06-01)
Pulsed-field gel electrophoresis (PFGE) characterization of 335 temporally and spatially matched clinical, bovine, and human Salmonella enterica subsp. enterica isolates revealed 167 XbaI PFGE patterns. These isolates were previously classified into 51 serotypes and 73 sequence types, as determined by multilocus sequence typing. Discriminatory power of PFGE (Simpson's index, D = 0.991) was considerably higher than that of multilocus sequence typing (D = 0.920) or serotyping (D = 0.913). Although 128 PFGE ty...
Detailed chromosome measurements and karyotype asymmetry of some Vicia (Fabaceae) taxa from Turkey
MARTİN, ESRA; Yildiz, Hatice Kubra; KAHRAMAN, AHMET; Binzat, Okan Kaan; EROĞLU, HALİL ERHAN (2018-01-01)
This study examined the detailed chromosome measurements and karyotype asymmetries of seven taxa in the genus Vicia. The taxa are V. articulata, V. cassubica, V. villosa subsp. villosa in sect. Cracca, V. noeana var. noeana, V. sativa subsp. sativa, V. peregrina in sect. Vicia and V. caesarea in sect. Ervum. V. cassubica, V. noeana var. noeana, V. sativa subsp. sativa, V. caesarea have 2n = 12 chromosomes. V. articulata, V. villosa subsp. villosa, V. peregrina have 2n = 14 chromosomes in somatic cells. Tota...
