CYP1A is known to play important roles in the metabolism, detoxification and bioactivation of carcinogens and other xenobiotics in animals including fish. In our laboratory, CYP1A1 was obtained in a highly purified form with a specific content of 15-17 nmol P450 per mg protein from liver microsomes of feral fish, leaping mullet (Liza saliens). Purified mullet CYP1A1 showed a very high substrate specificities for 7-ethoxyresorufin and 7-methoxyresorufin in a reconstituted system containing purified fish P450 reductase and lipid. In addition, effects of each individual components of the reconstituted system, i.e., CYP1A1 and P450 reductase on 7-methoxyresorufin O-demethylase (MROD) activity were studied. 7-ethoxyresorufin O-deethylase (EROD) activity was strongly inhibited by cl-naphthoflavone (ANF). At 0.5 and 2.5 mu M, ANF inhibited EROD activity by 90 and 98%, respectively. Mullet CYP1A1 did not catalyze monooxygenations of other substrates such as aniline, ethylmorphine. N-nitrosodimethylamine and p-nitrophenol. Antibodies produced against CYP1A1 orthologues in fish such as trout and scup showed strong cross-reactivity with the purified mullet CYP1A1. In addition, anti-L. saliens liver CYP1A1 produced in our laboratory inhibited both the EROD and MROD activities catalyzed by L. saliens liver microsomes but stronger inhibition was observed with EROD activity. On the other hand, anti-mullet CYP1A1 antibodies showed very weak cross-reactivity with two proteins (presumably CYP1A1 and CYP1A2) in 3MC-treated rat liver microsomes. Moreover, 3MC-treated rat liver microsomal EROD activity was weakly inhibited by the anti-L. saliens liver CYP1A1. These results strongly suggested that the purified mullet CYP1A1 is structurally, functionally and immunochemically similar to the CYP1A1 homologues purified from other teleost species but functionally and immunochemically distinct from mammalian CYP1A1. A biomechanical and spectroscopic study of bone from rats with selenium deficiency and toxicity
Turan, B; Bayari, S; Balcik, C; Severcan, Feride; Akkas, N (2000-06-01)
Selenium, being an essential mineral in the mammalian diet, is important in providing protection against oxidative damage. Numerous in vitro studies of selenium compounds reveal a very high correlation between catalytic activity of selenium compounds and toxicity. The present study was designed to investigate the effects of dietary selenium on the biomechanical properties of bone. New born rats of both sexes were fed with either a control, or a selenium- and vitamin E-deficient, or a selenium-excess and vit...
COMPARATIVE-STUDIES OF SHEEP LUNG AND LIVER NITROFURANTOIN REDUCTASE
İşcan, Mesude (Elsevier BV, 1993-10-01)
1. Nitrofurantoin reductase which catalyzes the bioactivation of nitrofurantoin was purified to electrophoretic homogenity from sheep liver and lung microsomes, with a yield of 15% and 35%, respectively. The specific activity of both reductases was found to be similar (140 nmol/min/mg protein). 2. The effects of nitrofurantoin and NADPH concentrations, pH, ionic strength, amount of enzyme and reaction period, on the enzyme activity were studied and the optimum conditions for maximum activity of purified fi...
IN-VIVO EFFECTS OF THE ANESTHETIC, BENZOCAINE, ON LIVER MICROSOMAL CYTOCHROME-P450 AND MIXED-FUNCTION OXIDASE ACTIVITIES OF GILTHEAD SEABREAM (SPARUS-AURATA)
ARINC, E; SEN, A (Elsevier BV, 1994-03-01)
Gilthead seabreams were exposed to benzocaine, 4-aminobenzoic acid ethyl ester, 57 mg/l in sea water for 3 min, daily, for 2 or 3 consecutive days. The fish were killed 20 hr after the last treatment. Benzocaine treatment for 2 or 3 days resulted in 57% and 67% inhibition of liver microsomal aniline 4-hydroxylase and ethylmorphine N-demethylase activities, respectively. The total cytochrome P450 content of fish liver microsomes was unaltered following the 2-day benzocaine treatment. However, additional 3 mi...
ANALYSIS OF PLASMIDS MEDIATING ANT (2'') AND AAC(3)-II BASED GENTAMICIN RESISTANCE IN CLINICAL ISOLATES OF ENTEROBACTERIACEAE
Kocabıyık, Semra (Elsevier BV, 1989-07-01)
The genetics of ANT(2″) and AAC(3)-II mediated gentamicin resistance in Gram-negative bacteria obtained from Hacettepe University Hospital in Turkey, was studied. The plasmid DNA analyzed by agarose gel electrophoresis revealed a heterogeneous plasmid population. Conjugal transfer appeared to be an efficient mechanism for the dissemination of resistance to gentamicin/tobramycin/kanamycin/streptomycin/chloramphenicol/cephalothin, which was the common resistance pattern in the population. Plasmids of 46, 60, ...
Phenolic compounds, carotenoids, and antioxidant capacities of a thermo-tolerant Scenedesmus sp. (Chlorophyta) extracted with different solvents
Bulut, Onur; Akin, Dilan; Sonmez, Cagla; Oktem, Aysegul; Yucel, Meral; Öktem, Hüseyin Avni (Springer Science and Business Media LLC, 2019-06-01)
The human body can fight against the adverse effects of chronic exposure to environmental pollutants and stress by consumption of a diet rich in antioxidants. Although fruits and vegetables are the predominant sources, alternative sources of antioxidants such as microalgae are also being explored. Here, we investigate the antioxidant capacity, total phenolic, flavonoid, and carotenoid contents of novel thermo-resistant green microalga Scenedesmus sp. ME02. This strain has previously been shown to have a hig...
