The phytochemical compounds are used mostly in medicine as alternative medicine. Excessive cholesterol is removed mainly through conversion to bile acids in mammals, cytochromes P450 initiate all quantitatively significant pathways of cholesterol metabolism and bile acid biosynthesis. This study was aimed to investigate the possible potency of medicinal plant Epilobium hirsutum L. (EHT) extract and its ingredient ellagic acid (EAT) on rat liver cholesterol and bile acid metabolizing enzymes. In addition to molecular studies, bioactive compound of EHT extracts was identified using Liquid Chromatograph-Mass Spectrometry (LC-MS) technique. The water extracts of EHT and EAT were injected intraperitoneally as 37.5 mg/kg and 20 mg/kg for 9 days, respectively. Then, in vivo effects of the EHT and EAT extract on rat liver cholesterol and bile acid metabolizing CYPs were analyzed by determining protein and mRNA expression levels using western blotting and qRT-PCR techniques, respectively. In addition, serum cholesterol level of animals was determined. EHT caused 0.6, 0.8 and 0.6 fold decrease in protein expression of CYP7A1, CYP8B1 and CYP7B1, respectively, whereas 1.3 fold increase in protein expression of CYP27A1 was observed. EHT caused 2.1, 1.4 and 4.1 fold decrease in mRNA expression of CYP7A1, CYP27A1, CYP7B1, respectively. However, mRNA expression of CYP8B1 was decreased 2.3 fold by EHT treatment. EAT led to 0.7, 0.8 and 0.8 fold decrease in protein expression of CYP27A1, CYP8B1, CYP7B1, respectively, but one fold increase of protein expression of CYP7A1. EAT caused 2.6, 1.7, 1.8 and 2.3 fold increase in mRNA expression of CYP7A1, CYP27A1, CYP8B1 and CYP7B1, respectively. EHT-treated and EAT-treated animals also showed a significant reduction in total cholesterol level compared to control animals. Based on doses used that applied in this study, EHT and EAT is quite safe and can be used for drug development without any toxicity.

Effect of gemcitabine and retinoic acid loaded PAMAM dendrimer-coated magnetic nanoparticles on pancreatic cancer and stellate cell lines
Yalcin, Serap; Erkan, Mert; Unsoy, Gozde; Parsian, Maryam; Kleeff, Jorg; Gündüz, Ufuk (2014-07-01)
Gemcitabine is an anticancer drug used in the treatment of different cancer types, including pancreatic ductal adenocarcinoma. The maximum tolerated dose in humans is restricted by its side effects on healty cells. Furthermore, the fibrotic stroma produced by the pancreatic stellate cells prevents effective delivery of chemotherapeutic agents providing a safe-haven for the cancer cells. This becomes more of a problem considering the short half-life of this drug. Magnetic nanoparticle-based targeted drug del...
Effects of human granulocyte-colony stimulating factor on fracture healing in rats
Bozlar, M; Aslan, B; Kalaci, A; Baktiroglu, L; Yanat, AN; Tasci, A (2005-08-01)
Objective: Granulocyte colony stimulation factor (G-CSF) is generally used to prevent and cure the neutropenia associated with chemotherapy and bone marrow transplantation. In addition to its effects on neutrophil function, G-CSF was found to have the characteristic of modulating the cytokines in the inflammatory response. Then, the question to answer is whether it has any effect on fracture healing and to what extent? In this study, we test the effects of G-CSF on the healing of tibia fracture in a rat model.
EFFECT OF TANNIC ACID ON GLUTATHIONE S-TRANSFERASE AND NAD(P)H: QUINONE OXIDOREDUCTASE 1 ENZYMES IN RABBIT LIVER AND KIDNEY
KARAKURT, SERDAR; Adalı, Orhan (2011-01-01)
Glutathione S-transferase and NAD(P)H: quinone oxidoreductase 1 enzymes have important roles in detoxification and also activation of a wide variety of chemicals. Moreover, resistance of tumor cells against chemotherapeutic agents has been implicated in GST activities. The aim of this study was to investigate the effect of polyphenolic compound tannic acid on rabbit liver and kidney glutathione S-transferase and NAD(P)H: quinone oxidoreductase 1 enzyme activities. Tannic acid was found to be a potent inhibi...
Effects of aromatic diboronic acid on thermal charateristics of polybenzoxazines based on phenol and aniline
Telli, Aysegul Hisar; Hacaloğlu, Jale (2018-11-01)
In this work, polybenzoxazines based on phenol and aniline were prepared by curing as-synthesized and purified monomers in the presence of aromatic diboronic acid, BDBA. Condensation reactions between BOH groups of diboronic acid and phenolic OH groups of benzoxazine took place during curing only when unreacted phenol, or low mass oligomers were present in the medium. Generation of fragments involving B-O-C groups and significant improvement in thermal characteristics were detected upon curing the as-synthe...

Epilobium hirsutum (EH) is a medicinal plant for treating various diseases. Despite its wide usage, there is no available information about its potential influences on drug metabolism. The present study was undertaken to determine the in vivo effects of EH on hepatic CYP2B, CYP2C, CYP2D, and CYP3A enzymes that are primarily involved in drug metabolism. Male Wistar rats were injected intraperitoneally with EH water extract (EHWE) and ellagic acid (EA) at a daily dose of 37.5 and 20 mg/kg, respectively, for 9...
