In vitro regeneration of Anatolian black pine [Pinus nigra Arnold subsp. pallasiana (Lamb.) Holmboe] from excised embryos

Adventitious buds and shoots were regenerated in vitro from the excised embryos of Anatolian black pine (Pinus niqra subsp. pallasiana) on Schenk and Hildebrand (SH), Gresshoff and Doy (GD), White, Gamborg (G), White (W) and Woody Plant Medium (WPM) containing 22.4-44.4 μM benzyladenin (BA) and 0.054 μM α-naphthalene acetic acid (NAA). BA concentrations ranging from 8.9 μM to 44.4 μM were adequate for growth and development of adventitious shoots. For root development, 2.7 μM NAA was the best. Among the tested culture media, White and WPM were the most suitable media for both in vitro adventitious shoot and root formation from excised tissues of Anatolian black pine embryos.

Eleven populations of black pine (Pinus nigra Arnold subspecies pallasiana) were studied at 17 polymorphic isoenzyme gene loci. The mean number of alleles per locus (A) was 1.6.
