Objectives: This study aims to investigate the effects of cholesterol content and cationic property of liposomes on immune response. Materials and methods: Liposomes containing high amounts of 3ß-[N-(N',N'-dimethylaminoethane)- carbamoyl] cholesterol hydrochloride (DC-cholesterol) were prepared and loaded with K- and D-type CpG oligonucleotide (CpG-ODN) via dehydration-rehydration (DRV) method. After splenocytes and peritoneal exudate cells (PECs) primed with lipopolysaccharide (LPS) was incubated either with free or liposomal CpG-ODN counterparts, supernatants were collected and used in cytokine (IFN-g, IL-12 and IL-1ß) ELISA. Additionally, supernatants of PECs primed with LPS and stimulated with liposomes containing different doses of DC-cholesterol were collected and used in IL-1ß ELISA assay. Results: Low-dose CpG-ODN loaded liposomal formulations induced higher immune activation than free CpG-ODN at the same dose. While high-dose liposomal CpG-ODN formulations decreased pro-inflammatory cytokine production in splenocytes, they increased the secretion of IL-1ß. Inflammasome activation was increased in a dose dependent manner when PECs primed with LPS were incubated with only liposomes. Varying lipid molar ratios of DC-Cholesterol containing liposomes increased IL-1ß production based on increasing lipid molar ratio. Conclusion: This study revealed that type and lipid ratio of liposomes may alter the cellular efficacy of the loaded immune-stimulatory agent and may initiate inflammasome activation.

Metabological engineering of Pichia Pastoris for extracellular thermostable glucose isomerase production
Ata, Özge; Çalık, Pınar; Özdamar, Tunçer H.; Department of Biotechnology (2012)
The aim of this study is to develop a metabolically engineered P. pastoris strain for production of an active extracellular thermostable glucose isomerase (GI) enzyme by using genetic engineering techniques. For this purpose, research program was performed in two sub-programs. In the first sub-program, xylA gene from Thermus thermophilus was amplified and inserted into pPICZα-A expression vector. Thereafter, this pPICZα-A::xylA vector was cloned into AOX1 locus in P. pastoris genome and expressed under alco...
In vitro induction of growth and development of common juniper(Juniperus communis L.) from shoot and bud explants
Koçer, Zeynep Ahsen; Kaya, Zeki; Department of Biotechnology (2005)
The objective of the study was to investigate the optimum conditions for in vitro regeneration of common juniper (Juniperus communis L.) by using indirect organogenesis approach. Throughout the study; callus induction, organogenesis, improved organogenesis and root induction experiments were performed sequentially. It was found that explant position, genotype, gender, treatments and sampling time had significant effects on callus induction rate in common juniper. The results of treatments indicated that IBA...
Extracellular recombinant human growth hormone production by pichia pastoris
Orman, Mehmet Ali; Çalık, Pınar; Department of Chemical Engineering (2007)
In this study, the effects of bioprocess operation parameters on recombinant human growth hormone (rhGH) production by P. pastoris were systematically investigated. In this frame, first, for the extracellular expression and purification of human growth hormone by recombinant P. pastoris the cDNA of hGH, fused with a polyhistidine tag and also fused with a target site for the Factor Xa protease in which cleavage produces a mature N- and C- termini of rhGH, was cloned into pPICZαA plasmid and the constructed ...
Biorefining of sugar-beet processing wastes by anaerobic biotechnology: waste stabilization and bioproduct formation
Alkaya, Emrah; Demirer, Göksel Niyazi; Department of Environmental Engineering (2008)
The main objective of this study was to investigate two of the possible exploitation routes of anaerobic digestion (acid-phase and methane-phase) for the treatment of sugar-beet processing wastes, while producing valuable biobased products. For this purpose, four sets of laboratory experiments were carried out in a stepwise fashion: First, in the biochemical methane potential (BMP) assay (Set-up 1) wastewater and beet-pulp were efficiently digested (63.787.3% COD removal and 69.689.3% VS reduction) in batch...
Recombinant therapeutic protease production by Bacillus sp.
Korkmaz, Nuriye; Çalık, Pınar; Department of Chemical Engineering (2007)
The first aim of this study is the development of extracellular recombinant therapeutic protease streptokinase producing Bacillus sp., and the second aim is to determine fermentation characteristics for streptokinase production. In this context, the signal (pre-) DNA sequence of B.licheniformis (DSM1969) extracellular serine alkaline protease enzyme gene (subC: Acc. No. X03341) was ligated to 5’ end of the streptokinase gene (skc: Acc. No. S46536) by SOE (Gene Splicing by Overlap Extension) method through P...
