Thalassemia, a genetic blood disorder, occurs as a result of deformations of hemoglobin structures. Patients with thalassemia develop iron overload from chronic blood transfusions and require regular iron chelation to prevent potentially fatal iron-related complications. Deferiprone is a commercially available drug used as iron chelator for the treatment of thalassemia but the very long-term effectiveness is not clearly known yet. Therefore, some studies were carried out to find effective alternative drugs or delivery methods for treatment of thalassemia. Controlled delivery, which offers safer, more convenient, and more effective means of administering actives, seems promising with this respect. Chitosan, a natural biopolymer produced from deacetylation of chitin, has a variety of promising pharmaceutical uses and is presently considered as a novel carrier material in drug delivery systems. In this study, chitosan microspheres having different degree of deacetylation (DDA) and containing Deferiprone were prepared by oil/water emulsion method and by crosslinking with gluteraldehyde. Particle size, SEM, and in vitro drug release analysis were performed. The average sizes of the prepared microspheres increased with increasing degree of deacetylation of chitosan and with decreasing crosslinking degree. In vitro drug release studies showed that, the release rate of Deferiprone increased as the crosslinking degree increased, contrary to the expectations. This is explained by the crystalline structure of lightly crosslinked chitosan which have ordered and dense structure causing slower release rate for Deferiprone compare to highly crosslinked structures. In the second stage of the study, chitosan films hardened with gluteraldehyde were prepared by film casting method. IR, DSC and mechanical analysis were performed. For the films with various crosslinking degrees, it was

Histone Deacetylase Inhibition Activity and Molecular Docking of (E )-Resveratrol: Its Therapeutic Potential in Spinal Muscular Atrophy

Spinal muscular atrophy is an autosomal recessive motor neuron disease that is caused by mutation of the survival motor neuron gene (SMN1) but all patients retain a nearly identical copy, SMN2. The disease severity correlates inversely with increased SMN2 copy. Currently, the most promising therapeutic strategy for spinal muscular atrophy is induction of SMN2 gene expression by histone deacetylase inhibitors. Polyphenols are known for protection against oxidative stress and degenerative diseases. Among our ...
Liposome-encapsulated panomycocin: preparation, characterization and in vitro determination of antifungal efficacy against dermatophytes

Dermatopytosis is one of the most seen superficial fungal infections in humans resulting mild to severe inflammations. Antifungal drugs are not effective in the treatment of dermatophytosis due to serious adverse effects and resistance developed by fungal pathogens. Thus, the development of safe, naturally derived and potent antifungal/antimycotic agents which do not result in resistance and elicit little or no side effects to mammalian cells with high selectivity to the fungal pathogens is a continuing nee...
Synthesis and surface modification studies of biomedical polyurethanes to improve long term biocompatibility
Aksoy, Eda Ayşe; Hasırcı, Nesrin; Department of Polymer Science and Technology (2008)
Thrombus formation and blood coagulation is a major problem associated with blood contacting products such as catheters, vascular grafts and artificial hearts. An intense research is being conducted towards the synthesis of new hemocompatible materials and mdifications of surfaces with biological molecules. In this study, polyurethane (PU) films were synthesized in medical purity from diisocyanate and polyol without using any other ingredients and the chemical, thermal and mechanical properties were charact...
Mammalian Telomeric DNA Suppresses Endotoxin-induced Uveitis

Telomeric regions of mammalian chromosomes contain suppressive TTAGGG motifs that inhibit several proinflammatory and Th1-biased immune responses. Synthetic oligodeoxynucleotides (ODN) expressing suppressive motifs can reproduce the down-regulatory activity of mammalian telomeric repeats and have proven effective in the prevention and treatment of several autoimmune and autoinflammatory diseases. Endotoxin-induced uveitis (EIU) is an established animal model of acute ocular inflammation induced by LPS admin...
Nilotinib significantly induces apoptosis in imatinib resistant K562 cells with wild-type BCR-ABL, as effectively as in parental sensitive counterparts

Chronic myeloid leukemia (CML) is a hematological malignancy characterized by high levels of immature white blood cells. CML is caused by the translocation between chromosomes 9 and 22 (which results in the formation of the Philadelphia chromosome) creating BCR-ABL fusion protein. Imatinib and nilotinib are chemotherapeutic drugs which specifically bind to the BCR-ABL and inhibit cancer cells. Nilotinib is more effective in this respect than imatinib. We have shown that nilotinib induces apoptosis in imatin...
