The concurrent employment of alpha-amino acid-derived chiral components such as aldehydes and alpha-iso-cyanoacetates, in a sequential Ugi reaction/cyclization two-step strategy, opens the door to the synthesis of three structurally distinct piperazine-based scaffolds, characterized by the presence of L-Ala and/or L-Phe-derived side chains and bearing appropriate functionalities to be easily applied in peptide chemistry. By means of computational studies, these scaffolds have been demonstrated to act as minimalist peptidomimetics, able to mimic a well defined range of peptide secondary structures and therefore potentially useful for the synthesis of small-molecule PPI modulators. Preliminary biological evaluation of two different resistant hepatocellular carcinoma cellular lines, for which differentiation versus resistance ability seem to be strongly correlated with well defined types of PPIs, has revealed a promising antiproliferative activity for selected compounds.

Preparation of silica coated cobalt ferrite magnetic nanoparticles for the purification of histidine-tagged proteins
Aygar, Gulfem; Kaya, Murat; Özkan, Necati; Kocabıyık, Semra; Volkan, Mürvet (2015-12-01)
Surface modified cobalt ferrite (CoFe2O4) nanoparticles containing Ni-NTA affinity group were synthesized and used for the separation of histidine tag proteins from the complex matrices through the use of imidazole side chains of histidine molecules. Firstly, CoFe2O4 nanoparticles with a narrow size distribution were prepared in an aqueous solution using the controlled co-precipitation method. In order to obtain small CoFe2O4 agglomerates, oleic acid and sodium chloride were used as dispersants. The CoFe2O4...

Poly-1,1,2,2-tetrakis(4-9H-carbazol-9-yl)phenyl)ethene P(TCP) was successfully synthesized by electrochemical oxidation of corresponding monomer, namely, 1,1,2,2-tetrakis(4-9H-carbazol-9-yl)phenyl)ethene (TCP) using dichloromethane as the solvent and tetrabutylammonium hexafluorophosphate (TBAPF(6)) as supporting electrolyte. Spectroelectrochemical properties of P(TCP) were investigated in situ recording the electronic absorption spectra of the polymer film coated on indium-tin oxide (ITO) at various potent...
The effect of crosslinking on thermal inactivation of cellulases
Bilen, J; Bakir, U (1998-04-22)
Cellulase complex was crosslinked by using three different crosslinkers, dimethyl suberimidate (DMS), dimethyl adipimidate (DMA) and dimethyl-3-3'-dithiobispropionimidate (DTBP) and their effects on thermal inactivation were investigated. These reagents are all homobifunctional bisimidoesters having different crosslinking distances and crosslink only amino groups to each other. No significant activity loss was observed due to crosslinking with none of the crosslinkers used. In general, DMA and DTBP crosslin...
Partial purification and characterization of neutral trehalase from commercial baker's yeast, Saccharomyces cerevisiae
Yarar, S; Hamamcı, Haluk; Bakir, U (2000-12-01)
The neutral trehalase of a commercial baker's yeast (S. cerevisiae) strain has been partially purified using ammonium sulfate fractionation and DEAE-cellulose column chromatography techniques. Trehalase was precipitated between 35-50% ammonium sulfate saturation and approximately 5-8 fold purification was achieved. The yeast cAMP-dependent protein kinase was also precipitated in the same fraction and these two proteins were separated by DEAE-cellulose column chromatography. Trehalase became totally inactive...
Use of purified and modified bentonites in linear low-density polyethylene/organoclay/compatibilizer nanocomposites
Seyidoglu, Tijen; Yılmazer, Ülkü (2012-05-05)
Polyethylene-based ternary nanocomposites were prepared with different clay structures, obtained by the modification of purified Resadiye bentonite as the reinforcement, a random terpolymer of ethylene, butyl acrylate, and maleic anhydride with the trade name Lotader3210 as the compatibilizer, and linear low-density polyethylene (LLDPE) as the polymer matrix in an intensive batch mixer. The quaternary ammonium/phosphonium salts used for the modification of bentonite were dimethyldioctadecyl ammonium (DMDA) ...
