Catalase was entrapped in thermally reversible poly (isopropylacrylamide-co-hydroxyethyl-methacrylate) (pNIPAM/HEMA) copolymer hydrogels. The thermoresponsive hydrogels, in cylindIl-rical geometry, were prepared in an aqueous buffer by redox polymerization. It was observed that upon entrapment, the activity retention of catalase was decreased between 47 and 14%, and that increasing the catalase loading of hydrogel adversely affected the activity. The kinetic behaviour of the entrapped enzyme was investigated in a batch reactor. The apparent kinetic constant of the entrapped enzyme was determined by the application of lMichaeiis-Menten model and indicated that the overall reaction rate was controlled by the substrate diffusion rate through the hydrogel matrix. Due to the thermoresponsive character of the hydrogel matrix, the maximum activity was achieved at 25 degrees C with the immobilized enzyme. The K-m value for immobilized catalase (28.6mM) was higher than that of free enzyme (16.5mM). Optimum pH was the same for both free and immobilized enzyme. Operational, thermal and storage stabilities of the enzyme were found to increase with immobilization. (C) 1999 Society of Chemical Industry.

Immobilization of yeast cells in several conducting polymer matrices
Balci, Z; Akbulut, Ural; Toppare, Levent Kamil; Alkan, S; Bakir, U; Yagci, Y (2002-01-01)
Immobilization of yeast cells (Saccharomyces cerevisiae) in different polymer matrices was performed by constant potential electrolysis. These matrices were polypyrrole (PPy); poly(methyl methacrylate)/polypyrrole (,PMMA/PPy) and thiophene-capped poly(methyl methacrylate)/ polypyrrole (TPMMA/PPy). The characterization of PMMA/PPy copolymer was achieved by Fourier-transform Infrared Spectroscopy (FT-IR), Differential Scanning Calorimetry (DSC) and Scanning Electron Microscopy (SEM). The invertase activity of...
Immobilization of invertase in conducting copolymers of 3-methylthienyl methacrylate
Çırpan, Ali; Toppare, Levent Kamil; Yagci, Y. (Elsevier BV, 2003-4)
Immobilization of invertase in conducting copolymer matrices of 3-methylthienyl methacrylate with pyrrole and thiophene was achieved by constant potential electrolysis using sodium dodecyl sulfate (SDS) as the supporting electrolyte. Polythiophene (PTh) was also used in entrapment process for comparison. Kinetic parameters, Michaelis-Menten constant, Km, and the maximum reaction rate, Vmax, were investigated. Operational stability and temperature optimization of the enzyme electrodes were also examined.
Immobilization of Invertase in Copolymer of 2,5-Di(thiophen-2-yl)-1-p-Tolyl-1H-Pyrrole with Pyrrole
Celebi, Selin; Ibibikcan, Esin; Kayahan, Senem; Yigitsoy, Basak; Toppare, Levent Kamil (Informa UK Limited, 2009-01-01)
Immobilization of invertase in conducting copolymer matrix of 2,5-di(thiophen-2-yl)-1-p-tolyl-1H-pyrrole with pyrrole (poly(DDTP-co-Py)) was achieved via electrochemical polymerization. Kinetic parameters, Michaelis-Menten constant, Km and the maximum reaction rate, Vmax were investigated. Operational stability and temperature optimization of the enzyme electrodes were also examined. Immobilized invertase reveals maximum activity at 50 degrees C and; pH 8 and pH 4 for two copolymer matrices. Although the sa...
Immobilization of invertase in functionalized copolymer matrices
Erginer, Reyhan; Toppare, Levent Kamil; Alkan, Selmiye; Bakir, Ufuk (Elsevier BV, 2000-10)
In this study, immobilization of invertase on functionalized polymer electrodes constructed with pyrrole-capped polyazotetrahydrofuran and polytetrahydrofuran-block-polystyrene copolymer matrices was performed. Immobilization in these enzyme electrodes was carried out by file entrapment of the enzyme in conducting polymer matrices during electrochemical polymerization of pyrrole. Sodium dodecyl sulphate was used as the supporting electrolyte in the preparation of enzyme electrodes. The effects of temperatur...
Immobilization of tyrosinase in polysiloxane/polypyrrole copolymer matrices
Arslan, A; Kiralp, S; Toppare, Levent Kamil; Yagci, Y (Elsevier BV, 2005-04-01)
Immobilization of tyrosinase in conducting copolymer matrices of pyrrole functionalized polydimethylsiloxane/polypyrrole (PDMS/PPy) was achieved by electrochemical polymerization. The polysiloxane/polypyrrole/tyrosinase electrode was constructed by the entrapment of enzyme in conducting matrices during electrochemical copolymerization. Maximum reaction rate (V-max) and Michaelis-Menten constant were investigated for immobilized enzyme. Enzyme electrodes were prepared in two different electrolyte/sol vent sy...
