Cis-(Z)-flupentixol dihydrochloride (FLU), a thioxanthene drug, is used in therapy of schizophrenia as well as in anxiolytic and depressive disorders. Since the action mechanism of FLU is not completely understood, the main objective of present study is to provide a detailed evaluation of flupentixol-phospholipid membrane interactions at molecular level. FLU-dipalmitoylphosphatidylcholine (DPPC) interactions in presence and absence of cholesterol (CHO) were investigated as a function of temperature. The changes in upper part of membrane were more pronounced than those in central part of membrane, as indicated by EPR and FTIR. FLU was proposed to incorporate into phospholipid membranes with its triple ring parallel to head group and its chain toward alkyl chain of phospholipids. According to DSC results, the incorporation of 10 mol% FLU into DPPC caused a shoulder in transition peak, suggesting the occurence of a phase separation, and formation of this new phase is still observable in presence of CHO. It is well known that, structure and dynamics of lipids have significant influence on the function of membrane bound proteins, and consecutively their actions. Based upon these, it was proposed that FLU may modify membrane associated receptors and transport proteins, which would form the basis of its clinical efficiency. (C) 2016 Elsevier Ireland Ltd. Effect of bilateral somatosensory stimulus on oscillatory brain activity and long term memory
Göktepe, Gizem; Özkurt, Tolga Esat; Department of Medical Informatics (2017)
A psychotherapy treatment called eye-movement desensitization and reprocessing (EMDR) is used to elicit traumatic memories in shorter time and decrease its negative effect by using bilateral saccadic eye-movements, auditory and tactile stimuli. As yet, there is no well-accepted result with regard to the effect of bilateral stimuli on long term memory. In this thesis, effect of bilateral somatosensory stimulus on oscillatory brain activity and long term memory was investigated. Accordingly, a novel face reco...
Effects of risperidone treatment on the expression of hypothalamic neuropeptide in appetite regulation in Wistar rats
Kursungoz, Canan; AK, MEHMET; Yanık, Tülin (2015-01-30)
Although the use of atypical antipsychotic drugs has been successful in the treatment of schizophrenia, they can cause some complications in the long-term use, including weight gain. Patients using these drugs tend to disrupt treatment primarily due to side effects. The atypical antipsychotic mechanism of action regulates a number of highly disrupted neurotransmitter pathways in the brains of psychotic patients but may also cause impairment of neurohorrnonal pathways in different brain areas. In this study,...
Detection of hypothalamic neuropeptite levels involved in appetite regulation in aripiprazole treatment /
Özdemir, Serhat; Yanık, Tülin; Department of Biotechnology (2014)
Disruption or malfunctioning of neurotransmitter pathways, especially serotonergic and dopaminergic pathways are closely related to psychotic disorders such as schizophrenia. Antipsychotic medications are becoming widespread; however, in long term treatments, these drugs lead to considerable amount of weight gain as a side effect. Metformin, an antidiabetic drug, has previously been used to treat antipsychotic induced weight gain and aripiprazole has been defined as a neutral antipsychotic drug in terms of ...
Effect of arachidonic acid on specific binding of [H-3]naloxone to opioid receptors
Apaydin, S; Öktem, Hüseyin Avni (2000-01-01)
The effects of arachidonic acid (AA) on binding of [H-3]naloxone to the agonist and antagonist configurations of opioid receptors were investigated in rat brain. Equilibrium binding parameters of the agonist and antagonist configurations of the receptors were evaluated from homologue displacement data in the presence or absence of AA. Addition of AA at a concentration of 0.6 mM (1.5 mu mole/mg of protein) reduced by 22% and 53% the maximal number of binding sites (B-max) respectively in the absence or prese...
Effect of gemcitabine and retinoic acid loaded PAMAM dendrimer-coated magnetic nanoparticles on pancreatic cancer and stellate cell lines
Yalcin, Serap; Erkan, Mert; Unsoy, Gozde; Parsian, Maryam; Kleeff, Jorg; Gündüz, Ufuk (2014-07-01)
Gemcitabine is an anticancer drug used in the treatment of different cancer types, including pancreatic ductal adenocarcinoma. The maximum tolerated dose in humans is restricted by its side effects on healty cells. Furthermore, the fibrotic stroma produced by the pancreatic stellate cells prevents effective delivery of chemotherapeutic agents providing a safe-haven for the cancer cells. This becomes more of a problem considering the short half-life of this drug. Magnetic nanoparticle-based targeted drug del...
