Sustained release of a bioactive agent from a tissue engineering scaffold is one of the most common strategies to improve regenerative potential of the construct. However, depending on the chemistry of the agent, achieving high enough loading and controlled release can be challenging depending on the scaffold materials. These shortcomings can be solved by novel scaffold design fabricated by appropriate techniques and materials for the target tissue. In this study, an electrospun scaffold was designed to improve osteogenic activity of cells and diatom silica frustules were used to sustain loading and controlled release of a hydrophilic molecule, melatonin. Fibrous scaffolds were produced via wet electrospinning of the polymer blend solution poly(hydroxybutyrate-co-hydroxyvalerate (PHBV)/poly(ε-caprolactone) (PCL) which contains melatonin loaded diatom frustules. In the 3D fiber matrix diatom frustules were covered with a polymer coat which successfully lowered melatonin release more than half through 7 days achieving a controlled release. Melatonin had a concentration dependent effect on ALP activity of cells, while scaffolds bearing melatonin loaded frustules have significantly improved ALP activity of Saos-2 cells. Developed scaffold system has successfully induced osteogenic activity by controlled melatonin delivery and silica nature of diatom frustules which hold potential use for bone tissue engineering.

With the use of a scaffold as support material, adequate number of cells, and bioactive molecules, tissue engineering applications intend to promote the regeneration of tissues or to replace failing or malfunctioning tissues/organs. In this study, electrospun 2D and 3D cellulose-based scaffolds were aimed to be produced with pullulan (PULL). Cellulose acetate (CA) and PULL powders in various ratios (80/20, 50/50, and 80/20) were dissolved in DMAc/DMSO solvent system and electrospun as either 2D or 3D forms....
Polymeric scaffolds for bioactive agent delivery in bone tissue engineering
Uçar, Şeniz; Hasırcı, Nesrin; Yılgör, Pınar; Department of Chemistry (2012)
Tissue engineering is a multidisciplinary field that is rapidly emerging as a promising new approach in the restoration and reconstruction of tissues. In this approach, three dimensional (3D) scaffolds are of great importance. Scaffolds function both as supports for cell growth and depot for sustained release of required active agents (e.g. enzymes, genes, antibiotics, growth factors). Scaffolds should possess certain properties in accordance with usage conditions. Wet-spinning is a simple technique that ha...
Diatom shell incorporated PHBV/PCL-pullulan co-electrospun scaffold for bone tissue engineering
Dalgıç, Ali Deniz; Karataş, Ayten; Tezcaner, Ayşen; Keskin, Dilek (Elsevier BV, 2019-07-01)
Tissue engineering can benefit from wide variety of materials produced by microorganisms. Natural origin materials often possess good biocompatibility, biodegradability with sustainable production by microorganisms. A phytoplankton, diatom, produces an amorphous silica shell that can be obtained by a cost efficient production process. Diatom shells (DS) are promising for bone tissue engineering since silicon enhances bone regeneration. Biocompatible and biodegradable biopolymers with microorganism origin ca...

As a promising approach in tissue engineering, decellularization has become one of the mostly-studied research areas in tissue engineering thanks to its potential to bring about several advantages over synthetic materials since it can provide a 3-dimensional ECM structure with matching biomechanical properties of the target tissue. Amniotic membranes are the tissues that nurture the embryos during labor. Similarly, these materials have also been proposed for tissue regeneration in several applications. The ...

Sadreddini, Sanaossadat; Evis, Zafer; Keskin, Dilek; Department of Biomedical Engineering (2023-1-23)
Recently, bioceramic/polymer composites have dragged a lot of attention for treating hard tissue damages using bone tissue engineering (BTE). In this context, it was aimed to develop fibrous composite poly(hydroxybutyrate) co (hydroxyvalerate)- polycaprolactone, PHBV-PCL, scaffolds containing different amounts of baghdadite (BAG) and Ba-doped BAG that can provide bone regeneration in the bone defect area and to investigate the effect of these scaffolds on the structural, mechanical, and biological propertie...
