In this study, effect of cold stress and recovery on the superoxide dismutase (SOD) activities and the expression levels of Cu/ZnSOD gene were investigated in two barley cultivars (Tarm-92-winter type, Zafer-160-spring type). Eight days old barley seedlings were subjected to two different cold stresses; chilling stress at 4°C for 1, 3, 7 days and freezing stress at -3°C and -7°C. Analyses were performed both on leaf and root tissues. The SOD activities and isozyme patterns were determined by Native PAGE activity staining technique. Relative RT-PCR was used for the transcript levels of Cu/ZnSOD gene. The SOD activities and expression levels of control and cold stressed plants were compared by densitometric analysis. Under chilling stress, the activities of Cu/ZnSODx and Cu/ZnSOD2 did not have any significant change in leaf and root tissues of both cultivars. However, the expression levels of Cu/ZnSOD gene were more variable than activity results. After -3°C freezing stress, the activities of SOD enzyme in leaf tissues of Tarm-92 increased significantly, however, these activities significantly decreased in leaves of Zafer-160. These results suggested that both cultivars were not affected by chilling stress in terms of SOD enzyme activities and expression levels. Furthermore, under freezing stress conditions, the increment of SOD activities and expression levels in Zafer-160 was higher than Tarm-92. In conclusion, the changes in SOD isozyme activities and expression levels may not be enough for understanding of the cold stress mechanism. Therefore, further studies have to be carried on other antioxidant enzyme systems.

Effect of cold stress on antioxidant mechanism of winter and spring type barley (hordeum vulgare L.) cultivars
Afşar, Nilüfer; Yücel, Ayşe Meral; Department of Biochemistry (2007)
In this study, effect of cold stress on physiology and biochemistry of two Turkish barley cultivars, winter type Tarm-92 and spring type Zafer-160, was studied. For chilling stress treatment, cultivars were exposed to +4 ºC for 1, 3 and 7 days, and for freezing stress application acclimated cultivars (+4 ºC for 3 days) were treated with -3 ºC and -7 ºC. After freezing stress treatment, a recovery period was applied for 4 days at 4 ºC. Following analyses were performed on leaf and root tissues: growth parame...
Effects of antimitotic agents on haploid plant production from unpollinated ovules of sugar beet (beta vulgaris l.)
Gürel, S.; Gürel, E.; Kaya, Zeki; Erdal, M.; Güler, E. (Informa UK Limited, 2003-01-01)
The effects of antimicrotubule agents on haploid embryo formation from unpollinated ovules of sugar beet (Beta vulgaris L.) were investigated. The antimitotic agent colchicine (at 100 and 150 mg/l) and trifluralin (at 5.0 mg/l) increased the frequency of haploid embryo formation whereas pronamide (at 76.9 and 128.2 mg/l) and trifluralin (at 3.4 mg/l) decreased. Ovules that were non-treated with antimicrotuble agents (i.e., ovules of the control treatment) produced higher percentages of haploid embryos (4.25...
Effect of Anaerobic Azo Dye Reduction on Continuous Sludge Digestion
Ozkan-Yucel, Umay Gokce; Gökçay, Celal Ferdi (Wiley, 2014-10-01)
Effect of continuous feeding of a reactive azo dye, reactive orange 107, and its hydrolyzed form (HRO107), on a conventional anaerobic digester was investigated in this study together with observation of change in microbial community. Laboratory-scale digesters were fed with waste activated sludge and azo dye for 575 days continuously. The influent concentrations of reactive azo dye were between 200 and 3200 mg/L. The digester performance was not adversely affected by azo dye and its reduction metabolites t...
Effects of Formaldehyde Inhalation on Zinc, Copper and Iron Concentrations in Liver and Kidney of Male Rats
ÖZEN, OĞUZ ASLAN; KUŞ, İLTER; Bakirdere, Sezgin; Sarsilmaz, Mustafa; YAMAN, MEHMET (Springer Science and Business Media LLC, 2011-05-01)
In the present study, adult Wistar albino male rats were exposed to formaldehyde at different periods (subacute and subchronic) and concentrations (5.0 and 10.0 ppm) in order to figure out the changes in the concentration of Zn, Cu and Fe. It was observed that the formaldehyde inhalation caused gradual decline of body weights in the experimental groups when compared with control groups. It was found that subacute (4-week) or subchronic (13-week) exposure to formaldehyde for rats may cause growth retardation...
Effects of ammonium ion, acetate and aerobic conditions on hydrogen production and expression levels of nitrogenase genes in Rhodobacter sphaeroides OU001
Akkose, Sevilay; Gündüz, Ufuk; Yucel, Meral; Eroglu, Inci (2009-11-01)
In the present study, expression levels of nitrogenase encoding nifH and control genes nifA and prrA were examined at different physiological conditions in Rhodobacter sphaeroides O.U.001. In addition to variations in expression levels, changes in hydrogen production and growth were also investigated in response to different concentrations of ammonium source, acetate and aerobic conditions.
