An analytical method was developed for the determination of thiols in biological samples. Reverse phase chromatography coupled to ICP quaduropole MS or Orbitrap MS was employed for the separation and detection of thiols. For the determination of total thiols, oxidized thiols were reduced using dithiothreitol (DTT). Reduction efficiencies for species of interest were found to be close to 100%. Reduced thiols were derivatized by p-hydroxymercuribenzoate (PHMB) and then separated on a C8 column. Optimization of the extraction, separation and detection steps of the HPLC-ICP-MS and HPLC-Orbitrap MS methods was carried out. Detection limits for cysteine, homocysteine, selenocysteine, glutathione, selenomethionine and cysteinyl-glycine were found to be 18,34,39,12,128 and 103 fmol, respectively, using HPLC-Orbitrap MS and 730, 1110, 440, 1110 and 580 fmol for cysteine, homocysteine, selenocysteine, glutathione, and cysteinyl-glycine using HPLC-ICP-MS. Contrary to expectation, the LODs and RSDs are higher for the HPLC-ICP-MS instrument, therefore HPLC-Orbitrap MS was used for the determination of thiols in yeast samples. Three different brands of baker's yeast and a selenized yeast were analyzed. The GSH and cysteine levels found in these samples ranged from 4.45 to 17.87 mu mol g(-1) and 0.61 to 1.32 mu mol g(-1), respectively.

Photocatalytic properties and characterization of praseodymium-doped titanium dioxide
DOĞU, Doruk; Karakaş, Gürkan (2018-01-01)
In this study, Pr-TiO2 samples were synthesized using a sol-gel method. The effects of the calcination conditions on the structure and photocatalytic activity were examined, and the results are compared with pure TiO2 samples. The XRD analysis indicated that the anatase structure is stabilized by Pr, and rutile phase formation is inhibited by the addition of Pr. It was also observed that Pr doping enhances the surface area and inhibits the crystal growth. Photoluminescence analysis with 325 nm excitation sh...
Speciation of Selenium in Supplements by High Performance Liquid Chromatography-Inductively Coupled Plasma-Mass Spectrometry
BAKIRDERE, Sezgin; Volkan, Mürvet; Ataman, Osman Yavuz (2015-06-13)
Anion and cation exchange high-performance liquid chromatography (HPLC) combined with inductively coupled plasma-mass spectrometry (ICP-MS) were used for speciation of selenium in supplements. All the parameters in the extraction, separation, and determination procedures were optimized. Recovery studies for the selenium species from the anion and cation exchange columns were performed and there were no analyte losses. Limits of detection for selenium(IV), selenium(VI), Se in selenomethionine, and Se in sele...
Speciation and preconcentration of inorganic tellurium from waters using a mercaptosilica microcolumn and determination by hydride generation atomic absorption spectrometry
Korez, A; Eroglu, AE; Volkan, Mürvet; Ataman, Osman Yavuz (2000-12-01)
A separation-preconcentration method utilising a mercapto-modified silica microcolumn was developed for the determination of trace amounts of Te(rv) and Te(VI) in waters by hydride generation atomic absorption spectrometry. Bottled drinking water, tap water and sea-water samples were spiked with sub-ng ml(-1) concentrations of Te(IV) and the spiked solutions were subjected to the mercapto-modified silica microcolumn separation and enrichment procedure. The spike recovery values varied between 86 and 114%. I...
Speciation of Arsenic in Fish by High-Performance Liquid Chromatography-Inductively Coupled Plasma-Mass Spectrometry

Arsenic speciation in fish was performed using isocratic elution with cation exchange column high-performance liquid chromatography and inductively coupled plasma-mass spectrometry detection. The separation of arsenobetaine and dimethylarsinic acid was performed in 12 min. DOLT-4 dogfish liver and NIST RM-50 albacore tuna certified reference materials were used to evaluate the accuracy for the determination of total arsenic in fish. In addition, reference materials were analyzed for arsenobetaine and dimeth...
Modeling and control studies for a reactive batch distillation column
Bahar, Almıla; Özgen, Canan; Department of Chemical Engineering (2007)
Modeling and inferential control studies are carried out on a reactive batch distillation system for the esterification reaction of ethanol with acetic acid to produce ethyl acetate. A dynamic model is developed based on a previous study done on a batch distillation column. The column is modified for a reactive system where Artificial Neural Network Estimator is used instead of Extended Kalman Filter for the estimation of compositions of polar compounds for control purposes. The results of the developed dyn...
