Although Camptothecin (CPT) has significant antitumor activity to lung, ovarian, breast, pancreas, and stomach cancers; lactone ring opening causes a reduction in cytotoxic activity and severe side effects in physiological conditions (pH: 7.4, 37 degrees C) due to decreased cell membrane binding, membrane diffusibility, and intrinsic potency against the topoisomerase target. Taken into consideration the targeted delivery advantage of biocompatible magnetic particles, in order to contribute to the removal of the handicap ( lactone ring opening), the current study investigated stability, release, and in vitro bioactivity of CPT in oleic acid (OA)-pluronic F127 (PL) coated iron oxide micro and nanoparticles. CPT encapsulated magnetite micro (similar to 10 mu m) and nano (similar to 30 nm) formulations were synthesized without changing their magnetic properties. Analysis of encapsulated CPT illustrated that over 99% of the lactone form was protected in the magnetic particle formulations and release of CPT from the magnetic micro and nanoparticles lasted for 9 and 4 days respectively. In addition, in a representative in vitro cytotoxicity study, CPT encapsulated OA-PL stabilized magnetite microparticles were approximately 65% more effective than free CPT to inhibit 100% cell growth on three different human liver carcinoma cell lines. Therefore, the OA-PL stabilized iron oxide formulation is ideal for achieving targeted and sustained CPT delivery in bioactive lactone form to overcome its handicaps in clinical conditions.

MTA-1 expression is associated with metastasis and epithelial to mesenchymal transition in colorectal cancer cells
Cagatay, Seda Tuncay; Cimen, Ismail; SAVAŞ, BERNA; Banerjee, Sreeparna (Springer Science and Business Media LLC, 2013-04-01)
Although metastasis associated protein 1 (MTA1) has been widely linked to tumor metastasis, the relevant mechanisms remain to be elucidated, especially in colorectal cancer (CRC). Here, we have investigated the link between MTA1, metastasis and epithelial-mesenchymal transition (EMT) in CRC. Eighteen normal colon tissues and 91 resected tumor samples were analyzed for MTA1 expression by immunohistochemistry (IHC). IHC indicated low or no nuclear MTA1 expression in the normal tissues and significantly higher...
Comparison of NAT1, NAT2 & GSTT2-2 activities in normal and neoplastic human breast tissues
Geylan-SU, YS; Isgor, B; Coban, T; Kapucuoglu, N; Aydintug, S; Iscan, M; Iscan, M; Güray, Nülüfer Tülün (American Society for Pharmacology & Experimental Therapeutics (ASPET), 2006-01-01)
In this study, arylamine N-acetyltransferases, NATs (E.C.2.3.1.5) and glutathione-S-transferase-T2-2, GSTT2-2 (E.C.2.5.1.18) enzyme activities in the breast tumor and surrounding tumor-free tissues of 22 female breast cancer patients with infiltrating ductal carcinoma were measured. The possible impacts of grade of malignancy, chemotherapy treatment, estrogen receptor status and menopausal status on all enzyme activities were evaluated. The results showed that, both NAT2 and GSTT2-2 display significant diff...
Acyl chain length and charge effect on Tamoxifen-lipid model membrane interactions
Bilge, Duygu; KAZANCI, NADİDE; Severcan, Feride (Elsevier BV, 2013-05-22)
Tamoxifen (TAM), which is an antiestrogenic agent, is widely used during chemotherapy of breast, pancreas, brain and liver cancers. In this study, TAM and model membrane interactions in the form of multilamellar vesicles (MLVs) were studied for lipids containing different acyl chain length and different charge status as a function of different TAM (1, 6, 9 and 15 mol%) concentrations. Zwitterionic lipids namely dipalmitoyl phosphatidylcholine (DPPC), and dimyristoylphosphatidylcholine (DMPC) lipids were use...
Anti-estrogen Resistance in Human Breast Tumors Is Driven by JAG1-NOTCH4-Dependent Cancer Stem Cell Activity
Simoes, Bruno M.; et. al. (2015-09-01)
Breast cancers (BCs) typically express estrogen receptors (ERs) but frequently exhibit de novo or acquired resistance to hormonal therapies. Here, we show that short-term treatment with the anti-estrogens tamoxifen or fulvestrant decrease cell proliferation but increase BC stem cell (BCSC) activity through JAG1-NOTCH4 receptor activation both in patient-derived samples and xenograft (PDX) tumors. In support of this mechanism, we demonstrate that high ALDH1 predicts resistance in women treated with tamoxifen...
Comparison of GST Isoenzyme Expression in Normal and Neoplastic Breast Tissue: Correlation with Clinical and Prognostic Factors
OĞUZTÜZÜN, SERPİL; İşcan, Mesude; ÖZHAVZALI, MÜZEYYEN; Sak, Serpil Dizbay (2009-01-01)
Glutathione S-transferases in breast tissue play an important role in the susceptibility to the mutagenic effects of chemical carcinogens and in the response of breast tumors to chemotherapy. In this study the immunohistochemical staining characteristics of glutathione S-transferase isoenzymes (alpha, mu, pi, and theta) were investigated in invasive duct carcinomas and in normal breast tissue of 43 patients. The relationships between the expression of the GST isoenzymes and some clinicopathological features...
