The reconstruction of skeletal muscle tissue is currently performed by transplanting a muscle tissue graft from local or distant sites of the patient's body, but this practice leads to donor site morbidity in case of large defects. With the aim of providing an alternative treatment approach, skeletal muscle tissue formation potential of human myoblasts and human menstrual blood derived mesenchymal stem cells (hMB-MSCs) on synthetic [poly(l-lactide-co-caprolactone), 70:30] scaffolds with oriented microfibers, human muscle extracellular matrix (ECM), and their hybrids was investigated in this study. The reactive muscle ECM pieces were chemically crosslinked to the synthetic scaffolds to produce the hybrids. Cell proliferation assay WST-1, scanning electron microscopy (SEM), and immunostaining were carried out after culturing the cells on the scaffolds. The ECM and the synthetic scaffolds were effective in promoting spontaneous myotube formation from human myoblasts. Anisotropic muscle patch formation was more successful when human myoblasts were grown on the synthetic scaffolds. Nonetheless, spontaneous differentiation could not be induced in hMB-MSCs on any type of the scaffolds. Human myoblast-synthetic scaffold combination is promising as a skeletal muscle patch, and can be improved further to serve as a fast integrating functional patch by introducing vascular and neuronal networks to the structure. Influence of co-culture on osteogenesis and angiogenesis of bone marrow mesenchymal stem cells and aortic endothelial cells
Pekozer, Gorke Gurel; KÖSE, GAMZE; Hasırcı, Vasıf Nejat (2016-11-01)
Co-culture of bone forming cells and endothelial cells to induce pre-vascularization is one of the strategies used to solve the insufficient vascularization problem in bone tissue engineering attempts. In the study, primary cells isolated from 2 different tissues of the same animal, rat bone marrow stem cells (RBMSCs) and rat aortic endothelial cells (RAECs) were co-cultured to study the effects of co-culturing on both osteogenesis and angiogenesis. The formation of tube like structure in 2D culture was obs...
Magnesium Oxide Nanocomposites for Improving Orthopedic Tissue Regeneration
Hıckey, Danıel; Sun, Lınlın; Ercan, Batur; Webster, Thomas (null; 2014-12-31)
Where ligaments meet bone, there exists a transitional region of heterogeneous tissue that is graded from hard, highly mineralized fibrocartilage at the bone interface to un-mineralized soft tissue at the ligament. This structure, called the enthesis, disperses stress concentrations that arise due to the vastly different mechanical properties of bone and ligaments. However, in the event that a ligament is injured and requires surgery, the tendon-to-bone insertion site (TBI) must be destroyed and is not eas...
Surface functionalized poly-lactic acid (PLA) scaffolds for bone tissue engineering
Monirizad, Mahsa; Keskin, Dilek; Ermiş Şen, Menekşe; Department of Engineering Sciences (2022-2)
The need for more effective tissue grafts for orthopedic applications is one of the main research areas of tissue engineering. In bone tissue engineering (BTE), scaffolds that can mimic bone tissue both from mechanical and biological perspectives are investigated mostly. In this study, it was aimed to develop a BTE scaffold that can mimic bone ECM, mechanical strength and cell biocompatibility in a single design and thus, various groups of scaffolds were characterized in terms of mechanical, biocompatibilit...
Biomechanics of the Gazi type pelvis external skeletal fixator
Korkusuz, Feza; Kaymak, O; Citipitioglu, E; Akkas, N (1997-01-01)
External skeletal fixation is an alternative method of treatment to conventional therapy and open surgical procedures in pelvis fractures. The appropriate type of frame and the configuration of the connecting bars of these fixators are under investigation. In the Gazi type pelvis external fixator (GPEF) which has been developed, a 70 degrees angulation is applied to the connecting bars of the anterior quadrangular frame. This configuration, which is expected to improve the stability of the posterior column ...
Tissue Engineered, Guided Nerve Tube Consisting of Aligned Neural Stem Cells and Astrocytes
Yucel, Deniz; Kose, Gamze Torun; Hasırcı, Vasıf Nejat (2010-12-01)
Injury of the nervous system, particularly in the spinal cord, impairs the quality of life of the patient by resulting in permanent loss of neurologic function. The main limitation in spinal cord regeneration is the lack of extracellular matrix to guide nerves for functional recovery of the transected nerve tissue. In the present study, a tissue engineered nerve tube was prepared by wrapping neural stem cells (NSCs) on aligned fibers using a micropatterned film with astrocytes aligned along the microgrooves...
